Source

Bat coronavirus RS4081 Spike Trimer Protein, His Tag (SPN-S522g) is expressed from human 293 cells (HEK293). It contains AA Gln 16 - Pro 1181 (Accession # ATO98120.1 (R653A, K954P, V955P)).

Predicted N-terminus: Gln 16

Molecular Characterization

This protein carries a polyhistidine tag at the C-terminus.

The protein has a calculated MW of 134.9 kDa. The protein migrates as 150-180 kDa when calibrated against Star Ribbon Pre-stained Protein Marker under reducing (R) condition (SDS-PAGE) due to glycosylation.

The protein is designed as a trimer.

Endotoxin

Less than 1.0 EU per μg by the LAL method / rFC method.

Purity

>90% as determined by SDS-PAGE.

>90% as determined by SEC-MALS.

Formulation

Lyophilized from 0.22 μm filtered solution in 0.1 M Sodium citrate, pH5.5 with trehalose as protectant.

Introduction de fond

The spike protein of bat coronavirus RS4081, a sarbecovirus related to SARS-CoV, mediates viral entry by facilitating attachment and membrane fusion. RS4081’s spike can support ACE2-independent, trypsin-dependent cell entry, highlighting alternative pathways for host infection and its potential relevance to zoonotic spillover and cross-species transmission studies.
