Background

Pertuzumab (brand name Perjeta), a recombinant humanized monoclonal antibody developed by Roche/Genentech, targets HER2’s extracellular dimerization domain. It is indicated for HER2-positive breast cancer (early neoadjuvant/adjuvant, metastatic) in combination with trastuzumab and chemotherapy. Its mechanism blocks HER2 heterodimerization, inhibiting downstream signaling and inducing apoptosis.
