Background

KLK2 encodes kallikrein-related peptidase 2, a highly active trypsin-like serine protease that selectively cleaves at arginine residues. It belongs to the kallikrein family and is primarily expressed in prostatic tissue, where it participates in proteolytic processing. KLK2 protein is found in the extracellular region, extracellular exosomes, and secretory granules. A central function of KLK2 is cleavage of pro-prostate-specific antigen into its enzymatically active form, placing it in zymogen activation and proteolysis. Its selective expression in the male reproductive system supports its role in reproductive tissue biology. Pathogenic relevance for KLK2 is reported in soft tissue sarcoma, and the gene is highly expressed in prostate tumor cells, where it has been described as a prognostic marker for prostate cancer risk.
