    Flow cytometric analysis of Human Jurkat cells staining with PE-Labeled Monoclonal Anti-Human CD43 Antibody, Mouse IgG1 (AS672) (Cat. No. FABm065-01) at 1:20 dilution (5 μL of the antibody stock solution corresponds to labeling of 1e6 cells in a final volume of 100 µL), compared with isotype control antibody. PE signal was used to evaluate the binding activity (QC tested).

Background

CD43, also designated Ly-48, leukosialin, sialophorin, leukocyte sialoglycoprotein and W3/13, is a large type I transmembrane glycoprotein enriched with O-glycosylation and sialylation sites. It binds to CD54 (ICAM-1), CD62E, CD169 (Siglec-1), CD328 (Siglec-7) and MHC Class I molecules. Widely expressed on T lymphocytes, thymocytes, monocytes, granulocytes, bone marrow stem cells, pre-B cells, activated B cells and plasma cells—but absent on resting peripheral blood B cells, red blood cells and nonhematopoietic cells—CD43 modulates leukocyte functions (bidirectional adhesion, T cell costimulation, apoptosis induction) via intercellular interactions. Enzymatic shedding of activated leukocyte surface CD43 may exert negative regulation on adaptive immune responses, making it a valuable target for flow cytometry analysis.
