Workflow

Storage
Keep the unopened kit stored at 2-8 °C. Avoid using the kit beyond its expiration date. The opened kit should be stored per components table. The shelf life is 30 days from the date of opening.

For each experiment, a standard curve needs to be set for each microplate, and the specific OD value may vary depending on different laboratories, testers, or equipment. The following example data is for reference only. The sample concentration was cal-culated based on the results of the standard curve. The minimum detectable con-centration of IFN-α2 is less than 150 pg/mL.
Sample Values
Cell Culture Supernatant: Human peripheral blood mononuclear cells (5 x 10⁶ cells/mL) were cultured in RPMI 1640 supplemented with 10% fetal bovine serum, 2 mM L-glutamine, 100 U/mL penicillin, and 100 μg/mL streptomycin sulfate. Cells were untreated or treated with 10 ug/mL of poly I:C in the presence of Lipofectamine 2000 for 24 hours. Subse-quent analysis involved quantitative measurement of IFN-α2 concentrations in collected culture supernatants using standardized immunoassays.

Intra-Assay Statistics
Ten replicates of each of three samples containing different IFN-α2 concentrations were tested in one assay. Acceptable criteria: CV<10%.

Inter-Assay Statistics
Three samples containing different concentrations of IFN-α2 were tested in inde-pendent assays. Acceptable criteria: CV<15%.

Recovery
Recombinant IFN-α2 was spiked into three human serum samples, and then analyzed. The average recovery of IFN-α2 for serum samples is 103.6%.
