Recombinant Laminin 521 offers a defined cell matrix with superior lot-to-lot consistency. It supports faster stem cell growth in homogenous monolayer compared to basement membrane extracts. In addition, it is widely used for in vitro feeder-free culture of various hPSCs while maintaining high quality.
Transferring your stem cells to Laminin 521 is straightforward, requiring no special adaptation process. Whether you’re transitioning from Matrigel, Vitronectin and other Laminin protein, we recommend adapting your stem cells over at least two passages and using adding Rock inhibitor (such as Y-27632). If adaptation proves challenging, consider using a higher concentration of Laminin 521 protein (e.g. 10 μg/ml). Once your stem cells are well adapted after several passages, you can gradually reduce the working concentration and cell density.
To passage iPSCs on Laminin 521-coated surfaces, first ensure that the coating is prepare according to the manufacturers’s instructions. Dissociate the cells using enzymes, like EDTA, Accutase, TrypLE Select. Typically, the incubation time for dissociation is approximately 5 to 6 minutes, though it may vary based on the specific cell line. Laminin 521 often results in tighter cell attachment compared to other matrix proteins, so using a lower working concentration of Laminin 521 may help reduce the detachment time.
Laminin proteins play a key role in stem cell expansion and differentiation due to their diverse tissue-specific expression . For hPSC culture, Laminin 521 and Laminin 511 are ideal for the maintaining high-quality stem cells. For differentiation into hepatic and neural cell, Laminin 111 and Laminin 121 are recommended. Laminin 211 supports differentiation into skeletal muscle, neural and cardiac cells while, Laminin 411 is suitable for endothelial and pancreatic cells differentiation.
