For academic customers, we recommend research grade Laminin 521, which is suited for stem cell research and high affordable compared to other commercial extracellular cell matrixes. GMP grade laminin 521 is designed for large scale manufacturing process of iPSC in the clinical stage, which is produced under strict GMP systems and comply with regulations from multiple countries.
Recombinant Laminin 521 is the defined cell matrix with good lot-to-lot consistency. It supports faster stem cell growth in monogenous monolayer compared to basement membrane extracts. It is widely used for in vitro feeder-free culture of multiple human pluripotent stem cells and maintain the high quality.
No more special adaption process is needed when you transfer your stem cells from other commercial extracellular matrix proteins, such as Matrigel, Vitronectin and other Laminin proteins to our Laminin 521 protein. We recommend adapting your stem cells by at least 2 passages and adding Rock inhibitor (such as Y-27632). If it is hard to adapt, the higher working concentration of Laminin 521 protein (such as 10 μg/ml) is recommended. After several passages, if your stem cells are well adapted, you can reduce the working concentration and cell density.
We have tried different dissociation enzymes, such as EDTA, Accutase, TrypLE Select to passage iPSC. Generally, the incubation time is about 5-6 minutes and will be longer depending on special cell lines. Meanwhile, stem cells coated with Laminin 521 often attach tighter compared to other matrix proteins. The lower working concentration of Laminin protein should be used to reduce the detach time.
Laminin proteins are crucial for stem cell expansion and defined cell differentiation based on the expression in different tissues in human body. For human pluripotent stem cell culture, Laminin 521 and Laminin 511 are recommended for the expansion of high-quality stem cells. Laminin 111 and Laminin 121 are used in hepatic and neural cell differentiation. Laminin 211 could support skeletal muscle, neural and cardiac cells differentiation. Laminin 411 is used in endothelial and pancreatic cells differentiation.
