TIGIT (T cell immununo receptor with lg and ITIM domains, récepteur immunitaire de lymphocyte T avec domaines Ig et ITIM) est une protéine point de contrôle immunitaire nouvellement découverte qui est exprimée sur les lymphocytes T et les cellules tueuses naturelles (NK). TIGIT est un récepteur inhibiteur qui module l'activité immunitaire en entrant en compétition avec les récepteurs activateurs CD226 (également connus sous le nom de DNAM-1) pour la fixation du ligand. Cet interrupteur marche/arrêt de TIGIT et DNAM-1 est parallèle à celui de CTLA4 & CD28, ce qui en fait une cible prometteuse pour la thérapie anti-tumorale.
CD155 (PVR) et CD112 (PVRL2 ou Nectine-2) sont des co-ligands de TIGIT et DNAM-1, tandis que CD113 (PVRL3 ou Nectine-3) se lie uniquement à TIGIT. L'affinité entre les différents ligands et récepteurs varie. La liaison entre TIGIT et CD155 est plus robuste que celle entre TIGIT et CD112 ou CD113.
