The minimum detectable concentration of IFN-γ is less than 5.309 pg/mL.
Cell Culture Supernatant – Human peripheral blood mononuclear cells (PBMCs) were cultured and stimulated using anti-CD3&CD28 magnetic beads and several concentrations of lipopolysaccharide (LPS) then detected IFN-γ secretion amount on Day 3 (72 hours).
High concentrations of human IFN-γ were added to different diluted substrates for gradient dilution to evaluate the linearity of the assay.
20 replicates of each of 3 samples containing different IFN-γ concentrations were tested in one assay. Acceptable criteria: CV<10%.
3 samples containing different concentrations of IFN-γ were tested in independent assays. Acceptable criteria: CV<10%.
IFN-γ was spiked into 3 human serum samples, and then analyzed. The average recovery of IFN-γ for serum samples is 93.5%, range is 85.2% - 105.2%.
